Design Notes

The exact sequence of the stop codon between gene III and GFP in J100287 is "TAA", with the second "A" also being the first base of the "ATG" start codon at the beginning of GFP. Therefore, two bases (the "TA" in the stop codon) had to be removed for the gene III and GFP sequences to be in the same reading frame.

The sequence for this part comes from J100287, which itself comes from the J100265 (pJC173b) plasmid that the Campbell synthetic biology lab at Davidson College received from the Liu lab at Harvard University.
