Design Notes
The exact sequence of the stop codon between gene III and GFP in J100287 is "TAA", with the second "A" also being the first base of the "ATG" start codon at the beginning of GFP. Therefore, two bases (the "TA" in the stop codon) had to be removed for the gene III and GFP sequences to be in the same reading frame.
